Overview

Acetylated, amidated analog of Selank, a tuftsin-derived heptapeptide; Russian research on the parent peptide reports anxiolytic and mild nootropic effects without sedation.

N-Acetyl Selank Amidate is a terminally modified analog of Selank, itself a synthetic analog of the immunopeptide tuftsin. Russian studies of the parent peptide report anxiolytic effects attributed to GABAergic and serotonergic modulation, plus enkephalinase inhibition and immunomodulatory (IL-6, interferon) effects, without the sedation or dependence of benzodiazepines. The acetylated-amidated analog has no published trials of its own; greater stability and potency are inferred from its chemistry and reported by users.

Benefits

Cognitive support, anxiolytic

Dosing reference

Research dose
Vendor/community range 250 to 500 mcg once or twice daily, intranasal; no published human or animal dose for this analog.
Range
250 to 500 mcg, 1-2x daily
Reconstitution
2 mL bacteriostatic water
Common vial sizes
5 mg

Intranasal per vendors. No pharmacology has been published for the acetylated, amidated analog; data for the parent Selank do not transfer automatically.

Mechanism

No pharmacology of the acetylated, amidated form is published; claims of better stability, absorption or potency are extrapolation. For unmodified Selank, Russian work reports inhibition of enkephalin-degrading enzymes (Zozulya et al., 2001), changes in GABAergic gene expression in rodent brain (Volkova et al., 2016) and raised hippocampal BDNF after intranasal dosing in rats (Inozemtseva et al., 2008).

Evidence

Anecdotal: no peer-reviewed data

  • No study of N-acetyl Selank amidate in any species has been published
  • Unmodified Selank was studied as an anxiolytic in a Russian trial in generalized anxiety disorder and neurasthenia (n=62) (Zozulia et al., 2008)
  • Unmodified Selank is reviewed as a heptapeptide anxiolytic with several proposed molecular targets (Vyunova et al., 2018)
  • Human-trial status: none for this analog. Unmodified Selank is a registered drug in Russia (Deigin et al., 2022); that registration does not extend to the modified form

Side effects

Nasal irritation, mild drowsiness, and occasional dizziness are the reported effects. Safety data for this modified analog specifically is limited; the available evidence comes from unmodified Selank in Russian trials.
