Overview

Peptide mimetic of neural cell adhesion molecule (NCAM) defined in patent filings; one published study tested it in rats and monkeys, and the memory literature belongs to its parent FGL.

FGL-S is a peptide mimetic of NCAM whose structure is defined in a patent family from ENKAM rather than in a peer-reviewed paper. It appears in one published study, which measured pharmacokinetics and emotional behavior for several NCAM mimetics in rats and non-human primates. The memory, FGFR1 and neuroprotection literature in circulation is for FGL, the parent 15-residue peptide, and the human phase 1 study was of FGL-L, a different peptide. No human data exist for FGL-S.

Benefits

Cognitive support, nerve regeneration

Dosing reference

Research dose
Vendor/community range 500 to 2000 mcg daily; no published human dose for FGL-S.
Range
500 to 2000 mcg, 1x daily
Reconstitution
2 mL bacteriostatic water
Common vial sizes
10 mg

Subcutaneous per vendors. The only human dosing data belong to the related FGL-L: single intranasal doses of 25, 100 and 200 mg in 24 healthy men (Anand 2007). They do not establish a dose for FGL-S.

Mechanism

Proposed by analogy: the parent FGL peptide mimics the NCAM fibronectin type III module that binds fibroblast growth factor receptor 1, and FGL-S is assumed to share that action. No mechanism study of FGL-S itself is published. The only FGL-S paper measured pharmacokinetics and emotional behavior in rats and non-human primates (Turner et al., 2019).

Evidence

Anecdotal: no peer-reviewed data

  • FGL-S was tested with other NCAM peptide mimetics for pharmacokinetics and emotional behavior in rats and non-human primates (Turner et al., 2019)
  • The structure of FGL-S is defined in ENKAM patent filings, not in a peer-reviewed paper
  • Rodent memory, LTP and neuroprotection findings in circulation used the parent FGL peptide, not FGL-S
  • Human-trial status: none for FGL-S. A different NCAM mimetic, FGL-L (FGLL), was given intranasally at 25 to 200 mg to healthy volunteers in a phase 1 tolerability and pharmacokinetic study (Anand et al., 2007)

Side effects

No human safety data; effects unknown beyond animal studies, where FGL peptides were tolerated in rodents at the doses tested. The exact identity of material sold under this name is unclear in the literature, which adds product risk.

Storage

Storage
Refrigerate 2-8°C (36-46°F)
Lyophilized
2+ years at room temp, 3+ years refrigerated
Reconstituted
28-30 days refrigerated

Pharmacology

Half-life
No human data. Rat and primate pharmacokinetics were reported for NCAM mimetics (Turner et al., 2019); an FGL-S value was not extracted in this audit
Bioavailability
Not established in humans; no published bioavailability figure for FGL-S
