Overview

Vial blend of CJC-1295 without DAC (Mod GRF 1-29) and GHRP-2 (pralmorelin), 5 mg each; GHRP-2 raised GH and appetite in human studies, and no study has tested the pair.

Vendors in the catalog sell this as CJC-1295 without DAC (Mod GRF 1-29) and GHRP-2, 5 mg each. Mod GRF 1-29 has no published study of its own, and its D-Ala2 substitution only extends the half-time of GHRH(1-29) to about 7 min in men (Soule 1994). GHRP-2 is a synthetic hexapeptide ghrelin-receptor agonist approved in Japan only as a single-dose diagnostic for GH deficiency (Chihara 2007); it sustained GH and IGF-1 over a 30-day infusion in older adults and increased food intake 36% in healthy men. IV GHRP-2 given with GHRH was synergistic in children (Pihoker 1995), the rationale for the pairing, but no study has tested this blend. FDA lists injectable GHRP-2 in category 2 for 503B outsourcing and CJC-1295 in the withdrawn-nomination table of its compounding safety-risk page.

Benefits

GH release (human, GHRP-2); increased food intake (human, GHRP-2 infusion); GH release by Mod GRF 1-29 inferred from related GHRH analogs; body-composition and recovery claims untested for the blend

Dosing reference

Research dose
200 to 500 mcg of blend per injection (100 to 250 mcg of each at the 5 mg/5 mg ratio), 1 to 3x daily: vendor/community range, no published human dose of the blend or of Mod GRF 1-29
Range
200 to 500 mcg, 1-3x daily
Reconstitution
2 mL bacteriostatic water
Common vial sizes
10 mg

Subcutaneous in community use, usually fasted. Human GHRP-2 doses: 1 mcg/kg or 100 mcg IV (diagnostic), 1 mcg/kg/h by continuous SC infusion for 30 days in older adults, and 0.3 to 3 mcg/kg/day SC in GH-deficient children. No human dose of Mod GRF 1-29 has been published. The 5 days on, 2 days off pattern is community convention.

Mechanism

Proposed, from component and class studies. Mod GRF 1-29 is a tetrasubstituted GHRH(1-29) analog acting on the pituitary GHRH receptor; its activity has not been measured directly. GHRP-2 activates the ghrelin receptor (GHS-R1a), raising GH within 60 min, and in swine it also raised ACTH and cortisol where ipamorelin did not (Raun 1998). GHRH-receptor and ghrelin-receptor agonists given together raise GH more than either alone, shown for IV GHRP-2 with GHRH in children (Pihoker 1995). GHRP-2's ghrelin-like action also fits the rise in food intake seen with continuous infusion (Laferrere 2005).

Evidence

Preclinical: animal or in vitro data only

  • D-Ala2 substitution, one of Mod GRF 1-29's four changes, raised the plasma half-time of GHRH(1-29) from 4.3 to 6.7 min in 10 normal men (Soule et al., 1994)
  • IV GHRP-2 1 mcg/kg in 10 children gave a predictable GH rise with a plasma half-life of 33 min (Pihoker et al., 1998)
  • 30-day continuous SC infusion at 1 mcg/kg/h sustained pulsatile GH and raised IGF-1 in 17 older adults (Bowers et al., 2004)
  • SC GHRP-2 infusion increased ad libitum food intake 36% in 7 healthy men (Laferrere et al., 2005)
  • Human-trial status: no study has tested the blend as sold, and none has tested Mod GRF 1-29 itself. GHRP-2 is approved in Japan only as a GH-deficiency diagnostic (Chihara et al., 2007); IV GHRP-2 plus GHRH, not Mod GRF 1-29, was synergistic in children (Pihoker et al., 1995)

Side effects

No human safety study of the blend or of Mod GRF 1-29 has been published. A 30-day continuous GHRP-2 infusion kept safety screening tests normal in 17 older adults (Bowers 2004). GHRP-2 raised ACTH and cortisol in swine (Raun 1998) and increased hunger in men. Commonly reported by users (anecdotal): water retention, hunger, tingling in the hands and joint aches. CJC-1295 with DAC, a different product, has a half-life of 5.8 to 8.1 days, so mixing up the two forms changes exposure a great deal. Injectable and nasal GHRP-2 is in category 2 of FDA's 503B interim policy (added 2023-09-29), and CJC-1295 is in the withdrawn-nomination table for 503A, as of 2026-04-22: https://www.fda.gov/drugs/human-drug-compounding/certain-bulk-drug-substances-use-compounding-may-present-significant-safety-risks. FDA cites serious adverse events with CJC-1295, including increased heart rate and a systemic vasodilatory reaction.

Pharmacology

Half-life
Mod GRF 1-29 not measured; D-Ala2-GHRH(1-29) ~6.7 min (human, IV). GHRP-2 ~33 min (children, IV bolus). The ~30 min figure for Mod GRF is community lore
Bioavailability
Not measured for Mod GRF 1-29. GHRP-2 raised GH after SC and intranasal dosing in humans; absolute bioavailability not reported. No data for the blend
